Studiomux (if it works for you) has an effects plugin that lets you route directly over lightning cable - no audio interface required. Works best with AB3. Only one audio stream, but you can chain audio effects in AB3.

Do you know of a guitar or an harware synth with lightnig connector?...? Never heard of any...
