Can i do this effect with spire & sylenth1 if i can, can someone explain how?

You can do this using gate sidechaining with long release.

You need a monophonic sound with a moderate release. First note is held and pitchbent down an octave. Then some short notes are played while the pitch wheel is moved up slowly. The release part of the envelope gives you those volume fadeouts that Youtube guy has drawn in by hand, and just... retrigger the same note while the pitch bends.
As Youtube guy hinted at, it's note-based, no fancy creative effects required.
As for the timbre of the note, just get something dissonant. Turn on unison to about 6 or so. Squarewaves an octave and a fifth apart run through distortion might be a good starting point. Mess around with ringmod, FM, sync, different waveforms, etc.
